Research Description

The team of Marcus Buschbeck at the Josep Carreras Leukaemia Research Institute is looking for a motivated Postdoctoral Researcher to work on the main research line of the lab aiming to understand the link between chromatin architecture and metabolism provided by histone variants.

Histone variants can replace replication-coupled histones in the nucleosome and endow chromatin with unique properties (Buschbeck and Hake, 2017, NRMCB). Three macroH2A histone variants are unique in containing a linker domain and macrodomain in addition to the histone-fold. The macrodomain of macroH2A1.1 is able to bind the NAD+ derived metabolite ADP-ribose and ADP-ribosylated proteins. As a consequence macroH2A1.1 affects cell metabolism through binding NAD+-dependent effector proteins (Posavec Marjanovic, Hurtado-Bagès et al., 2017, NSMB). Furthermore, macroH2A proteins have a yet not fully understood function in regulating three-dimensional chromatin architecture (Douet et al., 2017, JCS and Kozlowski, Corujo et al., 2018, EMBO Rep.).

We part from the hypothesis that macroH2A proteins are metabolic sensors on chromatin. Preliminary data in the lab suggests that the two other macroH2A proteins have evolved to bind other, yet unknown metabolites. We are looking for a motivated postdoc who is interested to take on the challenge to identify these metabolites and to dissect the functional relevance of metabolite-histone binding for gene regulation, cell fate and chromatin architecture. The project will build on previous results and will combine biochemical and biophysical methods, in part through established collaborations. We will further use cell culture models to dissect the molecular mechanism and analyze primary samples from KO mice.

The postdoc will incorporate into a balanced team of postdocs and PhD students and interact with other lab members working on related projects. In particular, the candidate is expected to closely interact with lab members that have generated a large amount of preliminary results.

The contract will be funded by the Ministerio de Ciencia e Innovación, bajo la convocatoria Retos Investigación de Proyectos I+D+i, nº de expediente RTI2018-094005-B-I00.

What we need

  • Technical expertise in cell culture methods, molecular biology and biochemistry.
  • Competitive CV with a track record of publications that will allow us to apply for competitive fellowships such as MSCA, EMBO and FEBS.
  • PhD title.
  • 3-6 years of previous experience.
  • High level of collaborative and communicative skills.
  • Good level of English speaking and writing skills (required). Spanish (helpful).
